Has an American Football team ever scored 1 point in a game?

No.

BUT

In 1948, the AAFC's (predecessor of the AFL) Brooklyn Dodgers played a pre-season game against the CFL's Montreal Alouettes in which the first half featured Canadian rules and the 2nd half featured American rules. The final score was 27-1, with the Dodgers beating the Alouettes. Presumably the 1 point was scored on a "rouge", whatever that is.

The only way this would occur in a non-Canadian rules game would be for the defensive team to get a 1 point safety after they gave up a TD. More details here.

You can simulate this in NCAA Football for Playstation or XBox. Just score a TD, then on the 2 point try, run all the way back into your own end zone and either get tackled there or step out of bounds. The score should end up 6-1! Unfortunately, it appears that this little quirk wasn't included in the latest version.

A team does not necessarily need to run all the way back on a kickoff attempt for a 1 point score. Also, an extra point can be blocked and then recovered outside of the endzone by the defense which then runs into the endzone for the safety.

A rouge is kicking the ball through the endzone except on a FG or PAT where the other team doesn't advance it back out, and kickoffs going out of bounds aren't counted either

The 1 point safety is one of my favorite quirks in American football rules. If it happened it would probably be a turnover on a PAT taken back not quite all the way and turned over again (defense gets 2 points for a conversion attempt returned, offense gets 1 point for a turnover that the defense can't get out of the endzone, the latter looks like making a kick in the box score and that's what happened in a college bowl game a few years ago)

See also the fair catch kick in NFL and high school rules, can attempt a FG from the spot of a fair catch. There's no snap or blockers at the line so you get several more yards effective range. Even so it generally only would be relevant at the end of a half on a short punt from backed up field position. I remember a Bills - Patriots game years ago late in the 2nd quarter where the Pats punter bounced it along the sideline so Buffalo couldn't use that rule.
